The novel offers a glimpse into the battles between light and darkness, and goodness and evil, as the young characters mature into adolescents on their journey. L'Engle modeled the Murry family on her own. Scholar Bernice E. Cullinan noted that L'Engle created characters who "share common joy with a mixed fantasy and science fiction setting. The book has inspired two film adaptations, both by Disney : a television film directed by John Kent Harrison ; and a theatrical film directed by Ava DuVernay.

The book begins by relating Meg's personal struggles at school and her inability to fit in with the crowd. This is also a problem for her younger brother Charles Wallace. Everyone thinks he is dumb, though both children are extraordinarily intelligent - indeed, Charles Wallace could easily be considered a genius. Meg Murry sits in her attic bedroom, freaking out partly because there's a storm, but more because she feels all wrong. Her teachers think her stupid, her friends think she's a baby, and she beat up a boy for talking smack about her younger brother. It doesn't help that her father has disappeared. Meg hates herself for being unable to hold in her feelings and act normal like everyone else. She goes downstairs to make some cocoa, and finds her younger brother Charles Wallace is expecting her, and already has some cocoa going. Charles Wallace is the sibling Meg feels closest to not only does it seem like he can read her mind sometimes, but he's also an oddball like her. Everyone thinks he's slow, but really he's smarter than most people.

A Wrinkle in Time is the story of Meg Murry, a high-school-aged girl who is transported on an adventure through time and space with her younger brother Charles Wallace and her friend Calvin O'Keefe to rescue her father, a gifted scientist, from the evil forces that hold him prisoner on another planet. At the beginning of the book, Meg is a homely, awkward, but loving girl, troubled by personal insecurities and her concern for her father, who has been missing for over a year. The plot begins with the arrival of Mrs. Whatsit at the Murry house on a dark and stormy evening. Although she looks like an eccentric tramp, she is actually a celestial creature with the ability to read Meg's thoughts. She startles Meg's mother by reassuring her of the existence of a tesseract--a sort of "wrinkle" in space and time.
